Magnesium is most abundant cations in our body. It is essential for many physiological process and pathway. In the body, one half percentage magnesium present in our bone. It is also present in small tissue. In the blood cell, magnesium are present in small amount. Magnesium decrease level are observed when diabetics, more amount alcohol, hyperthrodism, malabsorption, heart failure, liver failure and mycordial infection. Magnesium increase level are observed when renal failure ( Kidney ), vitamin d intoxication.


Method of Magnesium Test :-
Magnesium are estimate by Xylidyl blue method from human serum. Xylidyl blue method are mostly used method to estimate magnesium level in the human serum. It is quantitative method to estimate the amount of magnesium in the human body.


Principle of Magnesium Test :-
Magnesium are cations in the body. When magnesium are formed purple colour complex with Xylidyl blue in alkaline solution. The intensity of xylidyl blue and magnesium purple colour is directly proportional to the magnesium concentration.


Chemical reaction of magnesium :-

Magnesium ( Mg ) + Xylidyl blue regent
:-
Mg + Xylidyl blue Complex ( Purple Colour Complex )



Specimen collection :-
For the estimation of magnesium mostly red colour tube ( plain tube ) of green tube ( lithium heparin tube )  are used. Because xylidyl blue method required heparinized plasma. Don't collect patient blood sample in Eyhlene diamine tetra acetic acid tube ( purple colour tube ),  citrate oxlate ( grey colour tube ) or sodium citrate ( blue colour tube ).

Regent storage :-
Magnesium regent are store in refrigerator at 2 to 8 degree Celsius. Expiry date are checked before use the regent.


Requirements for blood collection :-
1. Red colour tube or green colour tube for collection blood sample.
2. Sterile single used Syringe
3. Dry cotton, spirit cotton or alcohol dipped cotton.
4. Tourniquet


Materials used for xylidyl blue method :-
1. Sterile glassware
2. Plastic or glass tube ( Sterile or contaminated free tube )
3. Micro Pipette
3. Sterile micro pipette tips
4. Instrument :- Auto lab manually machine, incubator


Procedure of magnesium test :-
1. Red colour tube or green colour tube are used to estimate the magnesium from the patient blood sample.
2. Collected blood sample tube are centrifuge in centrifugation machine for 5 min at 3000 RPM.
3. Separate serum are used for xylidyl blue method.
4. Magnesium regent are make normal for used test because it stored in refrigerator at 2 to 8 degree Celsius.
5. After magnesium regent are ready to use pipette out 1.0 ml ( 1000 micro litre ) in plastic or glass tube.
6. Add 10 micro litre serum sample in tube containing magnesium regent.
7. Mix well
8. Incubate for 5 min at room temperature.
9. After incubation, aspirate out immediately in auto lab machine.
10. Reading are note down in book.



Precaution :-
1. When blood collection of patient used hand gloves, mask.
2. Use single used syringe. Dispose in red colour bag.
3. Use sterile glassware or clean glassware.
4. Magnesium regent are stored in refrigerator at 2 to 8 degree Celsius. When test are done kept in refrigerator to maintain regent temperature.
6. Use sterile or new plastic or glass tube for mixing magnesium regent and serum sample of blood.
7. Care must be taken to avoid magnesium contamination.
8. If plastic or glass ware are used soaked first in dilute HCL or in cleaner and dispose in bag or container.
9. All materials are used in test must be proper and clean in condition.

:- When magnesium interact with xylidyl blue regent purple colour complex are form in alkaline solution. The intensity of purple colour complex between xylidyl blue and magnesium is directly proportional to the concentration of magnesium in human blood serum.

6. How many amount of blood sample are required for magnesium test ?
:- 3 to 5 ml blood are required to the test. But 10 micro litre serum sample are required for xylidyl blue method.

:- Normal range :- 1.7 to 2.2 mg/dl


8. What is normal magnesium level for a women ?
:- 1.7 to 2.2 mg/dl
